The gun is never seen closely. It might be a S&W American, a Schofield...
Remington 1866 Derringer
John Wayne carries one of these small pistols hidden, and uses it at the beginning of the movie
Winchester 1894
Wayne carries one of these rifles, with a large loop lever, just like the one he used in other films like True Grit, but he doesn't shoot it, and we never see it closely
